Programme and venue (3-days course)

A. Basic Principles of Flow Cytometry (Stefaan Verhulst) and Line up of applications in flow cytometry (Introduction by Karine Breckpot followed by presentation of individual researchers using flow cytometry in a particular application) (morning).
B. An introduction to compensations and different flow cytometers and their specificity (Angelo Willems) (afternoon).

A. Detection of selected cytokines in a murine tumor lysate using luminex kit for cytometer bead array analysis. Practical aspects in the Biochemistry laboratory and explanation of the measurements by the person in charge at the 3-laser flow cytometer with attention to the pitfalls in multiplex bead analysis (morning).
B. Purification of 4 different immune cells (cytotoxic T cells, NK cells, monocytes and pDCs) from the PBMC fraction in the pig. Practical aspects at the Laboratory of Veterinary Immunology, explanation of sorting process by the person responsible for the cell sorter and ‘tip and tricks’ to prepare an efficient sort (afternoon).

A. Detection of antigen-specific T cells by intracellular cytokine determination (ICS) (morning).
B. Practical aspects: performing protocol (fix / perm) + antibody staining, followed by flow cytometric analysis with extra attention to pitfalls in ICS (afternoon).
